Title : To study the effect of alcohol on amylase activity using uv spectrophotometer.

Abstract :

Enzyme amylase catalyses the first step in the digestion of starch which is a main source

 

of carbohydrate in the human diet. The study of enzyme amylase is significant from

 

research point of view as it breaks down starch to sugars by hydrolysis. The inhibitory

 

effects of alcohol on enzyme amylase was assessed in this study. The present study

 

revealed the effects of ethanol on amylase activity both qualitatively and quantitatively.

 

Enzyme activity was quantified by the amount of the coloured intermediate

 

produced, the concentration of which was determined by using a

 

spectrophotometer at 560 nm. Amylase action was measured  qualitatively by the

 

Lugol’s solution which reacts with starch to produce deep blue color, the

 

concentration of which was determined by measuring the rate of change of

 

absorbance with time. The results from both group ( control & experimental ) revealed

 

that the rate of amylase reaction was dependent on the presence of ethanol  . An overall

 

higher absorbance of the control solution was indicated and the R-squared

 

value of the experimental group was approximately half the R-squared value of the

 

control which followed  a very linear regression trend.
